TheCultrex®InVitroAngiogenesisAssayEndothelialCellInvasionKitwascreatedinanefforttoacceleratethescreeningprocessforcompoundsthatinfluencevascularendothelialcelldigestionofandinvasionthroughabasementmembraneextract(BME)layer.ThiskitoffersaflexIBLe,standardized,96wellhigh-throughputformatforquantifyingendothelialinvasion,invitro,inresponsetochemoattractantsand/orinhibitingcompounds.ThisassayemploysasimplifiedBoydenchamber-likedesignwithan8micronpolyethyleneterephthalate(PET)membrane.Sulforaphane[1-isothiocyanato-(4R)-methylsulfinyl)-butane],anaturallyoccurringcancerchemopreventiveagent,isprovidedasacontrolforinhibitionofinvitroendothelialcellinvasionthroughCultrex®BME.DetectionofcellinvasionisquantifiedusingCalcein-AM,which isinternalizedbythecells,andconvertedtotheacetomethylester(AM)moietybyintracellularesterases.FreeCalceinfluorescesbrightly,andthisfluorescencemaybeusedtoquantifythenumberofcellsthathaveinvadedusingafluorescenceplatereader.
