Description
TheCultrex®InVitroAngiogenesisAssayTubeFormationKitwasdevelopedfortheevaluation ofinducersandinhibitorsofendothelialcelltubeformation. Duringthecourseoftheassay,endothelialcellsareseededonathickgelofBasementMembraneExtractinthepresenceofangiogenesismodulatingagents. Ifthemediumispermissivetotheangiogenicresponse,cellswillselfassembleintotubenetworkswithinhours;ifthereareinhibitorsinthemedium,tubenetworkformationwillbeincompleteornonexistent.Sulforaphane[1-isothiocyanato-(4R)-methylsulfinyl)-butane],foundinbroccoliandothercruciferousvegetables,isanaturallyoccurringcancerchemopreventiveagent,andisprovidedasacontrolinhibitortovisualizethisprocess. EndothelialcellsmaybefluorescentlylabeledusingCalceinAM,andimagescapturedusingamicroscopecameramaybeanalyzetoquantifytheangiogenicresponse. Cultrex®InVitroAngiogenesisAssayKitprovidesacost-effectivemethodforinvestigationofProspectiveangiogenesisinhibitorsina96wellplateformat.
