TACS•XL®embodiesamorerobustapproachfortheinsitudetectionofapoptosisthanothermethods.TheTACS•XL®Kits arebasedonincorporationofbromodeoxyuridine(BrdU)atthe3’OHendsoftheDNAfragmentsthatareformedduringapoptosis.TheincorporationofBrdUbyTdTismoreefficientthaneitherbiotinylatedordigoxigeninlabelednucleotidesusedinotherterminaldeoxynucleotidyltransferasedUTPnickendlabeling(TUNEL)-basedassays.Thedetectionsystemutilizesabiotinconjugatedanti-BrdUantibodyandstreptavidin-horserADIshperoxidase.Thecombinationofantibodyspecificitywiththesignalenhancingpropertiesofbiotinstreptavidinresultsinprecisecellularlabelingandthehighestsignal-to-noiseratioobservedincompetitivetesting.
TheTACS•XL®BasicKitprovidesthereagentsnecessaryforroutinelabeling,includingtwopermeABIlizationreagents,labelingbuffersandreagents,Strep-HRPandTACS-Nuclease™reagentsforgeneratingpositivecontrolswithyourownsamples.It isidealforresearcherswhoselabsareequippedforstandardimmunohistochemicalproceduresinvolvinghorseradish-peroxidasewanttoadapttheassayforfluorescencedetection. Thekitdoesn’tnotcontainanyperoxydase-substrateorcounterstain. Add-onTACS•XL® BlueLabelModuleand TACS•XL® DABModule detectionmodules areavailableseparately.
